Gene expression changes caused by estrogen treatment of breast cancer cells that re-express ERalpha was investigated by infecting ER-negative MDA-MB-231 breast cancer cells for 24 h with recombinant adenovirus encoding full-length human ERalpha (Ad-ERalpha) or control vector (Ad-LacZ), and treating them with 0·01% ethanol (vehicle control) or 10-8 M 17beta-estradiol (E2). After 48 h of treatment, total RNA was isolated and used for transcript profiling on Affymetrix GeneChips. Three independent biological replicates of this experiment were carried out. 本研究针对重新表达雌激素受体α(ERalpha)的乳腺癌细胞经雌激素处理后诱发的基因表达变化展开探究。实验流程如下:将雌激素受体阴性的MDA-MB-231乳腺癌细胞分别以编码全长人ERalpha的重组腺病毒(Ad-ERalpha)或对照载体(Ad-LacZ)感染24小时,随后分别用0.01%乙醇(溶剂对照)或10^-8 M 17β-雌二醇(E2)进行处理。处理48小时后,提取总RNA并在Affymetrix基因芯片(Affymetrix GeneChips)上开展转录组谱分析(transcript profiling)。本实验共完成3次独立生物学重复。关键词:有序
