Analysis of intracellular inorganic element alterations to human pluripotent stem cells at gene expression level. The hypothesis tested in the present study was that changes in intracellular inorganic elements, like potassium, would change the pluripotent signaling network and influence differentiation cues. Results provide important information on the response of human pluripotent stem cells to tetraethylammonium- and salinomycin-based changes to intracellular potassium, such as specific up- or down-regulated genes governing cell proliferation, cell growth, differentiation, and development of specific cell lineages. Total RNA obtained from 48hr treatment with and without tetraethylammonium (TEA) and salinomycin (SAL), with and without changes in media by the addition of potassium chloride or sodium chloride. Study included 6 samples, repeated twice, for a total of 12 samples. Untreated samples in E8 TeSR media served as controls.
本研究针对基因表达水平下细胞内无机元素变化对人多能干细胞(human pluripotent stem cells)的调控效应展开分析。本研究所验证的科学假说为:细胞内无机元素(如钾离子)的改变会扰动多能性信号网络,并影响细胞分化的调控信号。研究结果揭示了人多能干细胞对基于四乙基铵(tetraethylammonium,TEA)与盐霉素(salinomycin,SAL)诱导的细胞内钾离子水平变化的应答特征,具体包括调控细胞增殖、细胞生长、细胞分化以及特定细胞谱系发育的特异性上调或下调基因。本次研究的总RNA样本取自经48小时处理的细胞:处理组分别施加四乙基铵(TEA)、盐霉素(SAL),或通过添加氯化钾、氯化钠改变培养基离子浓度,同时设置未进行上述处理的对照组。本研究共设置6组独立样本,每组重复2次,总计12个样本。以E8 TeSR培养基中培养的未处理细胞作为空白对照。
