SpoIIE deficient mutants in C. beijerinckii were generated using CRISPR-Cas9 to remove 2,4 kb within the gene coding sequence. Amongst the spoIIE mutant isolates, one could not produce solvents. The genome of this isolate was sequenced to confirm the spoIIE disruption and screen for SNPs.

本研究通过CRISPR-Cas9技术,敲除拜氏梭菌(C. beijerinckii)SpoIIE基因编码序列内2.4 kb的片段,成功构建SpoIIE缺陷突变体。在获得的SpoIIE突变分离株中,有一株无法合成溶剂类产物。随后对该分离株的基因组进行测序,以验证SpoIIE基因的敲除情况,并筛选单核苷酸多态性(Single Nucleotide Polymorphisms,SNPs)。
