Lateolabrax japonicus Transcriptome or Gene expression

Lateolabrax japonicus is one of the main marine aquatic fish species, and is popularly cultured in East Asia due to its highly commercial value. In this research, RNA-Seq analysis of the L. japonicus were carried out to identify reproduction and growth related genes at L. japonicus ovary of different tree ages by Illumina sequencing technology. In total, 334,388,688 high quality reads were obtained in four libraries, i.e., four-year-old vary (4th_Ovary), three-year-old vary (3rd_Ovary), two-year-old vary (2rd_Ovary) and one-year-old vary (1st_Ovary), and de novo assembled into 101,860 Unigenes with an average unigene length of 879 bp. A total of 30,142 unigenes (29.59%) were annotated in the public databases: Nr database (Nr), Swiss-Prot, Kyoto Encyclopedia of Genes and Genomes (KEGG), and Cluster of Orthologous Groups of proteins (COG), based on sequence similarity. Comparative analysis revealed, 35,749, 43,657 and 36,819 differentially expressed genes (DEGs) were identified in three comparisons (4th_Ovary VS 3rd_Ovary, 4th_Ovary VS 2rd_Ovary and 4th_Ovary VS 1st_Ovary), in total 24,295 DEGs were different expressed in 4th_Ovary. Enrichment and pathway analysis of the DEGs was also carried out to excavate the candidate genes related to reproduction and growth. The present study enriches the genomic resources of L. japonicus and provides fundamental information for further studies.
日本花鲈(Lateolabrax japonicus)是主要的海洋经济鱼类之一,因其极高的商业价值,在东亚地区被广泛养殖。本研究采用Illumina测序技术,对不同养殖年龄的日本花鲈卵巢组织开展RNA测序(RNA-Seq)分析,以鉴定与繁殖及生长相关的功能基因。本研究共构建四个测序文库,分别为4龄卵巢(4th_Ovary)、3龄卵巢(3rd_Ovary)、2龄卵巢(2nd_Ovary)及1龄卵巢(1st_Ovary),总计获得334,388,688条高质量读段(reads)。经从头组装(de novo)得到101,860条单基因(Unigene),单基因平均长度为879 bp。基于序列相似性比对,共有30,142条单基因(占比29.59%)在公共数据库中获得注释,涵盖Nr数据库(Nr)、Swiss-Prot数据库、京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes, KEGG)及直系同源蛋白簇(Cluster of Orthologous Groups of proteins, COG)。对比分析结果显示,在三组对照组合(4龄卵巢vs3龄卵巢、4龄卵巢vs2龄卵巢、4龄卵巢vs1龄卵巢)中,分别鉴定得到35,749、43,657及36,819个差异表达基因(differentially expressed genes, DEGs),总计有24,295个差异表达基因在4龄卵巢样本中呈现差异表达。本研究对差异表达基因开展富集分析与通路分析,以挖掘与繁殖和生长相关的候选基因。本研究丰富了日本花鲈的基因组资源,可为后续相关研究提供基础数据支撑。
