Experiments have been performed as described in Materials and methods. Raw bioluminescence recordings are detrended by a 24-hour moving average and analyzed via the ChronoStar software. Results of the nonlinear fitting by ChronoStar are depicted in the last 6 columns of each data block. Data where the bioluminescence after transduction and selection of U-2 OS cells is low compared to average (red font) as well as those where the error of the nonlinear curve fit is larger than 0.3 (red shaded cells) are sorted out for the further analysis in S11 Fig. Note that sheet 1 contains results for shRNA constructs targeting EIF2S1, while sheet 2 contains results for the nonsilencing constructs. 本实验依照材料与方法部分描述的实验方案开展。原始生物发光记录经24小时移动平均法完成去趋势处理后,采用ChronoStar软件进行分析。ChronoStar软件输出的非线性拟合结果展示于每个数据块的最后6列。U-2 OS细胞经转导与筛选后测得的生物发光信号低于平均值的数据(以红色字体标注),以及非线性曲线拟合误差大于0.3的数据(以红色填充单元格标注),均被剔除,未纳入S11图的后续分析。请注意:第1张工作表包含靶向EIF2S1的短发夹RNA(short hairpin RNA, shRNA)载体构建体的实验结果,第2张工作表则包含非沉默对照载体构建体的实验结果。本数据集为XLSX格式。
