The peak energy used was 120kV, and 2 second exposures were corrected with offset and gain images. 样本采用Metscher(2009)的方法,使用碘-碘化钾(IKI)染色剂染色三个月。显微计算机断层扫描(Micro-CT,简称uCT)数据采集使用Feinfocus 10-160kV透射X射线源与Perkin Elmer XRD0822型1百万像素平板X射线相机,扫描峰值电压设为120kV,2秒曝光图像通过偏移校正图像与增益校正图像完成校正。采集得到的曝光数据通过OctopusV9软件重建为tif stack格式图像序列,最终体素尺寸为86.9微米。
