The goal of this study was to compare NGS-derived murine myometrial progenitor cell transcriptome profiling (RNA-seq) of an animal model of uterine fibroid tumorigenesis in rats exposed during uterine development to an endocrine-disrupting chemical, diethylstilbestrol (DES) versus vehicle (VEH) control, unexposed animals, to determine the pathways and biological processes, whose gene expression may be altered as a result of the exposure. Overall design: Examination of gene expression in exposed vs. unexposed murine myometrial progenitor cells; myometrial progenitor cells isolated from 5 exposed animals were pooled and compared to that of the same cells isolated from 5 unexposed animals
本研究旨在对比子宫发育阶段暴露于内分泌干扰物己烯雌酚(diethylstilbestrol,DES)的大鼠子宫肌瘤发生动物模型,与未暴露的赋形剂(VEH)对照组大鼠的下一代测序(Next-Generation Sequencing,NGS)来源鼠类子宫肌层祖细胞转录组谱(RNA-seq),以明确暴露事件引发的基因表达改变所涉及的信号通路与生物学过程。实验整体设计:比较暴露组与未暴露组鼠类子宫肌层祖细胞的基因表达;从5只暴露大鼠中分离得到的子宫肌层祖细胞被混合为一组,与从5只未暴露大鼠中分离得到的同种细胞进行对照分析。
