Replicative DNA polymerase usage at genome wide.

We attmpt to establish a novel sequencing approach to characterise usage of replicative DNA polymerases in hct116 cells, which is derived from human colon carcinoma. This approach allows us to determine the roles of DNA polymerase delta and epsilon in lagging strand and leading strand DNA synthesis, respectively in genome-wide scale.

本研究旨在建立一种新型测序方法，以表征源自人结肠癌的HCT116细胞中复制型DNA聚合酶的使用模式。该方法可在全基因组规模下，分别确定DNA聚合酶δ（DNA polymerase delta）与ε（DNA polymerase epsilon）在滞后链与前导链DNA合成中的作用。
