Bovine tissue and blood contamination from bullets by LC-MS/MS

We have measured the proteomes of different major bovine organs in multiple replicate. Before the actual sample preparation, organs were penetrated with metal bullets experimentally. The biological material on these projectiles was then "on-surface" digested using trypsin and analyzed using LC-MS/MS. Further experiments have been done testing the influence of different metal types, shooting and the contamination with human blood.

本研究对多种主要牛源器官的蛋白质组开展了多次重复检测。在正式样本制备前，我们先通过实验方式用金属子弹穿透各器官样本。随后对这些弹体上附着的生物材料采用胰蛋白酶进行表面原位消化（on-surface digestion），并通过液相色谱-串联质谱（LC-MS/MS）完成分析。后续我们还增设了额外实验，用以评估不同金属材质、射击操作以及人血液污染对实验的影响。
