In order to determine how gene expression in nodules changes diurnally and to compare them with gene expression in roots, we used RNAseq to quantify gene expression in nodules and roots of plants transferred from a diurnal 12/12h light-dark photoperiodic cycle to constant light. We sampled nodules and roots immediately after the transfer and then after every 3 hrs for the first day, then every 6 hrs for the second day (13 time-points- 0h, 3h, 6h, 9h, 12h, 15h, 18h, 21h, 24h, 30h, 36h, 42h, 48h)

为明确根瘤中基因表达的昼夜动态变化规律,并将其与根系的基因表达水平进行对比,我们采用RNA测序(RNAseq)技术,对从12小时光照/12小时黑暗的标准昼夜光周期环境转移至持续光照条件下的植株的根瘤与根系进行基因表达定量分析。我们于植株完成转移后即刻采集根瘤与根系样本,随后在首日每3小时采样一次,次日每6小时采样一次,共计13个采样时间点:0h、3h、6h、9h、12h、15h、18h、21h、24h、30h、36h、42h、48h。
