To examine the cell and molecular transitions between endothelial (E), hemogenic endothelial (HE), and intra-arterial clusters (IAC) cells, and the heterogeneity of hematopoietic stem and progenitor cells (HSPCs) within the IACs, we profiled ~40,000 cells from the caudal arteries (dorsal aorta, umbilical, vitelline arteries) of embryonic day (E) 9.5 to 11.5 mouse embryos by single-cell RNA sequencing (scRNA-Seq). We identified a developmental bottleneck in endothelial to hematopoietic transition (EHT) and heterogeneity within the IAC population.
为探究内皮细胞(Endothelial,E)、生血内皮细胞(Hemogenic Endothelial,HE)与动脉内簇(Intra-arterial Clusters,IAC)之间的细胞及分子转变过程,以及动脉内簇内造血干细胞和祖细胞(Hematopoietic Stem and Progenitor Cells,HSPCs)的异质性,我们对胚胎发育第9.5至11.5天小鼠的尾动脉(包含背主动脉、脐动脉及卵黄动脉)中的约4万个细胞开展了单细胞RNA测序(Single-Cell RNA Sequencing,scRNA-Seq)分析。本研究鉴定出内皮向造血转化(Endothelial to Hematopoietic Transition,EHT)过程中的发育瓶颈,以及动脉内簇群体内部的异质性。
