KLF7, a member of the KLF family, is an evolutionarily conserved zinc finger-containing transcription factor. Previous studies demonstrated that KLF7 possesses diverse regulatory functions related to embryogenesis, cell growth, proliferation, and differentiation. Our results reveal that there was an increased abundance of KLF7 in OSM-treated HaCaT cells. Mechanistically, our results showed that OSM induces epidermal keratinocyte differentiation through phosphorylation of STAT5, which binds to the promoter and activates KLF7 transcription. Overall design: Examination of KLF7 bingding sites in Keratinocyte HaCaT cells.

KLF7是Krüppel样因子(Krüppel-like Factor, KLF)家族的成员,是一种进化保守的含锌指结构的转录因子。既往研究表明,KLF7具备与胚胎发生、细胞生长、增殖及分化相关的多种调控功能。本研究结果显示,经制瘤素M(Oncostatin M, OSM)处理的HaCaT角质形成细胞中,KLF7的丰度显著升高。机制分析结果表明,OSM通过磷酸化信号转导与转录激活因子5(STAT5)诱导表皮角质形成细胞分化,磷酸化的STAT5可结合至KLF7的启动子区域并激活其转录。整体实验设计:检测HaCaT角质形成细胞中的KLF7结合位点。
