To define the cellular composition and spatial architecture of the tumor micoenvironment, we combined single-cell RNA-sequencing, spatial transcriptomics, and multiplexed ion-beam imaging from 10 patient cutaneous squamous cell carcinoma tumors and site-matched normal skin SCC cell lines were infected with a CRISPR sgRNA library of 334 genes (and 136 non-targeting sgRNA controls) and grown either as xenograft tumors in immunocomprised mice or in vitro, followed by sequencing of sgRNAs

为解析肿瘤微环境的细胞组成与空间结构,我们整合了来自10例皮肤鳞状细胞癌(cutaneous squamous cell carcinoma)患者肿瘤及位点匹配正常皮肤样本的单细胞RNA测序(single-cell RNA-sequencing)、空间转录组学(spatial transcriptomics)与多重离子束成像(multiplexed ion-beam imaging)数据。同时,我们将鳞状细胞癌细胞系感染含334个基因的CRISPR单引导RNA(sgRNA)文库(配套136个非靶向sgRNA对照),分别于免疫缺陷小鼠体内构建异种移植瘤或在体外培养,最终完成sgRNA测序。
