Chenopodium ambrosioides is a perennial herb native to tropical America and is reported to be a Mn/lead (Pb) hyperaccumulator. In China, C. ambrosioides is well known as a common invasive alien species and can flourish in HM-contaminated habitats. In our previous study, we assessed the genetic and epigenetic variation of 21 C. ambrosioides populations in China, which covered a wide geographical distribution and different HM-contaminated habitats. Two different genetic clusters were detected in the eastern and southwestern populations. We also found that epigenetic variation might contribute to local metalliferous habitat adaptation. In this study, we assessed the molecular basis of three different Mn-tolerant C. ambrosioides populations. Among them, XC and CS belong to one genetic cluster and are distributed in Mn-contaminated and uncontaminated habitats, respectively, while WH belongs to the other cluster and is distributed in uncontaminated habitats. By integrating physiological, transcriptomic and epigenomic data, we investigated i) the differences in transcriptional and physiological responses to Mn stress among three C. ambrosioides populations and ii) the effect of epigenetic variation on Mn tolerance in C. ambrosioides.
土荆芥(Chenopodium ambrosioides)是一种原产于热带美洲的多年生草本植物,据报道为锰(Manganese, Mn)/铅(Lead, Pb)超富集植物(hyperaccumulator)。在中国,土荆芥是广为人知的常见外来入侵物种(invasive alien species),可在重金属(Heavy Metal, HM)污染生境中茁壮生长。在前期研究中,我们对中国境内21个土荆芥种群(population)的遗传与表观遗传变异(genetic and epigenetic variation)进行了评估,这些种群覆盖了广泛的地理分布范围与多种不同的重金属污染生境类型。研究在东部与西南种群中检测到两个不同的遗传簇(genetic clusters),同时发现表观遗传变异可能助力土荆芥适应本地含重金属的生境。本研究针对3个耐锰土荆芥种群的分子基础展开了评估。其中,XC与CS隶属于同一遗传簇,分别分布于锰污染生境与未污染生境;WH则隶属于另一遗传簇,分布于未污染生境。本研究整合生理、转录组(transcriptomic)与表观基因组(epigenomic)数据,开展了两项核心研究:①3个土荆芥种群对锰胁迫(Mn stress)的转录与生理响应差异;②表观遗传变异对土荆芥耐锰性的影响。
