本发明公开了一种海伦兜兰原球茎的低温
保存培养基及低温保存繁殖方法。它是将无菌播
种获得的原球茎转移至低温保存培养基中,在合
适的条件下培养,然后将低温保存的原球茎转移
至恢复生长培养基中,在合适的条件下,恢复生
长分化出小苗,经生根壮苗培养获得完整植株。
本发明的方法具有保存成本低、操作简单、植物
材料复苏快、生活力高等优点,在需要进行种苗
的大规模生产时,能在短期内获得大量的试管
苗,对于解决快繁生产急用及种质资源的保存也
均具有极其重要的价值。
This invention discloses a low-temperature preservation medium for protocorms of Paphiopedilum helenae and a low-temperature preservation and propagation method. Specifically, protocorms obtained via sterile sowing are transferred to the low-temperature preservation medium and cultured under suitable conditions. Subsequently, the cryopreserved protocorms are transferred to a regeneration medium, and cultured under appropriate conditions to resume growth and differentiate into seedlings. Complete plants are finally obtained through rooting and seedling strengthening cultivation. The method of this invention boasts multiple advantages including low preservation cost, simple operation, rapid resuscitation of plant materials and high viability. When large-scale production of seedlings is required, a large number of test-tube plantlets can be obtained in a short period. It is of extremely important value for addressing the urgent needs in rapid propagation production and the preservation of germplasm resources.
