We report the effects of VapC21 overexpression on Mycobacterium tuberculosis H37Rv strain. The total RNA was isolated from M. tuberculosis harboring either pTetR-int or pTetR-Int-vapC21 the expression was induced with 50 ng/ml anhydrotetracycline for 24 hrs. We report the effect of deletion VapC21 on the transciptional profile of Mycobacterium tuberculosis Erdman strain. For RNA-seq analysis, total RNA was isolated from mid-log phase culture of either parental or △vapC21 mutant strain. Overall design: Experiments were repeated thrice for each overexpression strain and twice for each deletion strain.

本研究报道了VapC21过表达对结核分枝杆菌(Mycobacterium tuberculosis)H37Rv菌株的影响。实验分别从携带pTetR-int或pTetR-Int-vapC21的结核分枝杆菌中提取总RNA,以50 ng/ml脱水四环素诱导表达24小时。本研究同时报道了VapC21基因缺失对结核分枝杆菌Erdman菌株转录谱的影响。针对RNA测序(RNA-seq)分析,总RNA分别从亲本菌株或△vapC21突变菌株的中期对数生长期培养物中提取。实验设计:每株过表达菌株的实验均重复3次,每株缺失菌株的实验均重复2次。
