We analyzed the effect of diet-induced obesity on liver gene expression in wild-type mice and in mice lacking p110alpha in hepatocytes. Samples were collected from mice either fed or fasted. Mice lacking p110 alpha in hepatocytes were created by mating the floxed-p110α mouse strain (LWT) with C57BL/6J albumin-Cre transgenic mice to obtain albumin-Cre+/−p110αflox/flox mice, i.e., p110αhep-/- mice (LKO). Twelve-week-old p110αhep+/+ (LWT) and p110αhep-/- (LKO) mice were fed a HFD with 60% calories from fat (D12492, Research Diet) or a control (“CTRL”) diet (D12450J, Research Diet) during 12 weeks. All mice had free access to food and water. At the end of experiment, mice were fasted at Zeitgeber Time (ZT) 16 (with ZT0 being when the light is turned on and ZT12 when light is turned off) for 24h or fed ad libitum prior sacrifice. Mice were killed at ZT16.

将12周龄的p110αhep+/+(LWT)与p110αhep-/-(LKO)小鼠分别喂食脂肪供能占比60%的高脂饮食(HFD,D12492,Research Diet)或对照(CTRL)饮食(D12450J,Research Diet),造模周期为12周。所有小鼠均可自由获取食物与饮水。
实验结束时,在处死前,小鼠于授时因子时间(Zeitgeber Time, ZT)16开始禁食24小时,或保持自由进食;其中ZT0为光照开启时刻,ZT12为光照关闭时刻。所有小鼠均于ZT16时刻被处死。
