By using the AG1478 inhibition model and RNA-seq analysis, we discovered the importance of EGFR in self-renewal and pluripotency through regulating cell cycle and the expression of pluripotency factors. As a cell surface receptor conveying exogenous stimulations, EGFR deficiency led to cell cycle arrest, and impaired self-renewal and pluripotency. These findings will improve our understanding of the molecular mechanisms driving the self-renewal and pluripotency of mESCs.

本研究通过构建AG1478抑制模型并开展RNA测序(RNA-seq)分析,明确了表皮生长因子受体(EGFR)通过调控细胞周期及多能性因子表达,在干细胞自我更新与多能性维持中的核心作用。作为介导外源刺激的细胞表面受体,EGFR缺失会引发细胞周期阻滞,进而损害自我更新能力与多能性。上述研究结果将深化我们对驱动小鼠胚胎干细胞(mESCs)自我更新与多能性的分子机制的认知。
