High-throughput small molecule screening revealed that high concentrations of cytoplasmic calcium ([Ca2+]c) were linked to dormancy of HSCs.To clarify molecular difference between [Ca2+]chigh and [Ca2+]clow cells, we performed RNA-Seq analysis using [Ca2+]chigh and [Ca2+]clow cells. Transcriptomes of [Ca2+]high HSCs and [Ca2+]low HSCs were generated by deep sequencing, in 7 replicates, using Next-Seq system.

高通量小分子筛选实验显示,高浓度胞质钙离子([Ca²⁺]c)与造血干细胞(hematopoietic stem cells, HSCs)的休眠状态存在关联。为厘清[Ca²⁺]c高表达细胞与[Ca²⁺]c低表达细胞间的分子差异,我们对两类细胞开展了RNA测序(RNA-Seq)分析。通过NextSeq测序系统,对[Ca²⁺]c高表达造血干细胞与[Ca²⁺]c低表达造血干细胞进行了7次生物学重复的深度测序,获取了二者的转录组数据。
