The objective of the present study is to explore the role of human MSC derived adipocytes transduced with/without lenti adipo-NaKtide or lenti adipo-scrambled NaKtide in experimental uremic cardiomyopathy. We investigated the pathophysiological changes in recipient mice that underwent partial nephrectomy surgery, as a model of uremic cardiomyopathy, and implanted with NaKtide transfected human MSC derived adipocytes. We aimed to demonstrate that adipocyte specific NaKtide reprograms the adipocyte phenotype and transplantation of these metabolically healthy adipocytes in diseased condition exhibits improved systemic and cardiovascular function, through intra-organ crosstalk. RNA was extracted from Heart, Kidney and Visceral adipose tissues of SCID mice (12 weeks old, male) in each group; SCID-Sham, SCID-PNx, SCID-ControlCells-PNx, SCID-Lenti-NaKtideCells-PNx, SCID-Scrambled-NaKtideCells-PNx, and sequenced in a 2x50 base-pair design on an Illumina 1500 sequencer.
本研究旨在探讨转染/未转染脂肪细胞靶向NaKtide慢病毒(lenti adipo-NaKtide)或乱序对照NaKtide慢病毒(lenti adipo-scrambled NaKtide)的人源间充质干细胞(Mesenchymal Stem Cell, MSC)衍生脂肪细胞在实验性尿毒症性心肌病中的作用。本研究以接受部分肾切除术以构建尿毒症性心肌病模型的受体小鼠为研究对象,分析其在移植经NaKtide转染的人源间充质干细胞衍生脂肪细胞后的病理生理变化。本研究旨在证实:脂肪细胞特异性NaKtide可重编程脂肪细胞表型;在疾病状态下移植这类代谢健康的脂肪细胞,可通过器官内串扰改善机体整体系统功能与心血管功能。从各组12周龄雄性重症联合免疫缺陷(Severe Combined Immunodeficiency, SCID)小鼠的心脏、肾脏及内脏脂肪组织中提取RNA,各组包括SCID假手术组(SCID-Sham)、SCID部分肾切除术组(SCID-PNx)、SCID部分肾切除术+对照细胞组(SCID-ControlCells-PNx)、SCID部分肾切除术+lenti adipo-NaKtide转染细胞组(SCID-Lenti-NaKtideCells-PNx)、SCID部分肾切除术+lenti adipo-scrambled NaKtide转染细胞组(SCID-Scrambled-NaKtideCells-PNx);随后采用Illumina 1500测序仪,以2×50碱基对(2x50 base-pair)双端测序方案完成转录组测序。
