We developed a ptpn6 knock-out zebrafish and found a disruption in the early development of the hematopoietic system. Marcrophage subpopulation size en numbers of macrophage progenitors differ between mutants and siblings. Overall design: 5dpf zebrafish embryos were dissociated and FACS sorted. Macrophages (mpeg) were sorted in 384 well plates.
本研究构建了ptpn6基因敲除(ptpn6 knock-out)斑马鱼模型,并发现在造血系统早期发育过程中存在异常。突变体与同窝对照的巨噬细胞亚群规模及巨噬细胞祖细胞数量均存在差异。整体实验设计:将受精后5天(days post fertilization, dpf)的斑马鱼胚胎解离,通过荧光激活细胞分选(fluorescence-activated cell sorting, FACS)进行分选;将以mpeg标记的巨噬细胞分选至384孔板内。
