Propagation of primary CRCs into organoids. Exome sequencing of CRC organoids

We propagated in vitro primary CRCs as 3D primary cultures. CRC cells grew indefinitely as epithelial structures lacking stromal cells called tumor organoids. We expanded organoids from 8 primary CRCs. Exome sequencing analysis confirmed that these organoids displayed distinct combinations of mutations in the main driver pathways.

我们在体外对原代结直肠癌（Colorectal Cancer, CRC）组织进行3D原代培养传代扩增。结直肠癌细胞可作为不含基质细胞的上皮样结构实现无限增殖，这类结构被称为肿瘤类器官（tumor organoids）。我们从8例原代结直肠癌组织中成功扩增获得此类肿瘤类器官。经外显子组测序（Exome sequencing）分析证实，这些类器官在主要驱动通路中呈现出各异的突变组合模式。
