We analysed gene expression profiles of D2.OR-EGFP-shControl and D2.OR-EGFP-shEpB6 cells isolated from lungs Overall design: D2.0R-EGFP-shControl (1x106 cells/mouse) or a mix of D2.0R-EGFP-shEphB6 cells (shEphB6#31, #34, #35) were injected in the tail vein of nude athymic BALB/c mice. After two weeks, lungs were harvested and digested into a single-cell suspension as described above. CD45-/EGFP+ cells were sorted directly into lysis buffer and total RNA was extracted with RNeasy Plus Micro Kit (Qiagen) following the manufacturer’s instructions.

本研究分析了从肺部分离得到的D2.OR-EGFP-shControl与D2.OR-EGFP-shEpB6细胞的基因表达谱。实验整体设计:将D2.0R-EGFP-shControl细胞(1×10⁶个细胞/小鼠)或D2.0R-EGFP-shEphB6细胞混合株(shEphB6#31、#34、#35)经尾静脉注射至无胸腺裸BALB/c小鼠体内。两周后,收取小鼠肺部并按照前述方法消化为单细胞悬液;将CD45⁻/EGFP⁺细胞直接分选至裂解缓冲液中,随后依照制造商说明书,使用RNeasy Plus Micro Kit(Qiagen)提取总RNA。
