Lactic acid producing bacteria were isolated from pickled mustard greens collected in Hanoi, Vietnam, using modified MRS agar. A 1 g sample was serially diluted in saline and plated, then cultivated in MRS broth at 37 degrees Celsius. P7 cells were harvested, washed, and their genomic DNA extracted with the GeneJET kit, checked via agarose gel electrophoresis, and quantified by NanoDrop. The P7 whole genome was sequenced using the Illumina NovaSeq platform with the objective of determining its complete genome sequence.
本研究从越南河内采集的腌芥菜样本中,采用改良MRS琼脂(modified MRS agar)分离产乳酸细菌。称取1g样品置于生理盐水中进行梯度稀释后涂布平板,随后于37℃下在MRS肉汤(MRS broth)中培养。收集P7菌株菌体并洗涤后,采用GeneJET试剂盒(GeneJET kit)提取其基因组DNA,通过琼脂糖凝胶电泳(agarose gel electrophoresis)进行质量检测,并利用NanoDrop进行浓度定量。本研究采用Illumina NovaSeq平台对P7菌株的全基因组进行测序,以明确其完整基因组序列。
