To better understand the contribution of basophils and fibroblast IL-4 signaling on type 2 skin inflammation, we conducted bulk-RNA seq on from normal and MC903 treated Isotype Cntrl + Mar1 and Il4rafl/fl + Il4raΔPdgfra models respectively. MC903 treated or vehicle control neck skin was harvested from WT Isotype control and MAR1 injected mice, Il4rafl/fl and, Il4raΔPdgfra mice (n=1 mice per sample). Briefly, skin samples were harvested, snap frozen, and cryosectioned. RNA was isolated and purified from each sample using the Quick-RNA Micro Prep Kit (Zymo Research). 为深入探究嗜碱性粒细胞与成纤维细胞的IL-4信号通路在2型皮肤炎症中所发挥的作用,我们分别对正常对照组、经MC903处理的同型对照+Mar1模型小鼠,以及经MC903处理的Il4rafl/fl + Il4raΔPdgfra模型小鼠的样本开展了批量RNA测序(bulk-RNA seq)。我们从野生型同型对照小鼠、注射MAR1的小鼠、Il4rafl/fl小鼠以及Il4raΔPdgfra小鼠处采集了颈部皮肤样本,其中部分样本经MC903处理,其余为赋形剂对照处理组,每个样本对应1只小鼠。简要实验流程如下:收集皮肤样本后迅速冷冻并进行冰冻切片;使用Zymo Research公司的Quick-RNA Micro Prep Kit完成各样本RNA的分离与纯化;文库构建及测序工作由诺禾致源(Novogene)完成。
