Extent of microbial contamination in assembled contigs of Hypsibius dujardini genome of Boothby et al. is confirmed by mapping sequence reads obtained from a single individual of H. dujardini Z151 (same strain) using ultra low input sequencing protocol using Thruplex kit (Rubicon Genomics). Mapping was performed using BWA, and mapping statistics are calculated using Qualimap software. <br>As shown in the report, nearly 7000 contigs show coverage &lt; 1, including 11 longest contigs.

本研究采用鲁比肯基因组学公司(Rubicon Genomics)Thruplex试剂盒(Thruplex Kit)的超低起始量测序流程,对Boothby等人研究中杜氏高生熊虫(Hypsibius dujardini)Z151品系的单个个体进行测序并获取测序读段(sequence reads),将该读段比对至其基因组组装重叠群(assembled contigs),从而确认了该组装序列中的微生物污染程度。比对工作采用BWA(Burrows-Wheeler Aligner)软件完成,比对统计数据则通过Qualimap软件计算得到。<br>正如报告所示,近7000个组装重叠群的覆盖度(coverage)低于1,其中包含11个最长的重叠群。
