Gene expression profiling reveals a potential role of Luteolin in LPS induced depression model LPS depression induced mice were orally treated with luteolin (10 mg/kg body weight) once per day during 8 consecutive days. Microarray gene expression was conducted for isolated mice Neural Stem Cells (NSCs) . Two mice brain were used for each expermient. The Microarray gene expression was conducted on NSCs and hipocampus from LPS depression induced depression mice (LPS group), LPS depression induced depression mice treated with luteolin(LPS+L group), Normal mice (PBS group) and Normal mice treated with luteolin (PBS+L). RNA was extracted using Isogen (Nippon Gene Co. Ltd., Toyama, Japan). The integrity of RNA was quantified using NanoDrop 2000 spectrophotometer (Thermo Scientific, Wilmington, DE, USA). RNA samples were prepared for gene expression profiling analysis with GeneChip® 3' Expression Arrays using 3' IVT PLUS Reagent Kit (Affymetrix Inc., Santa Clara, CA, USA). One hundred ng of total RNA from each sample was used to generate amplified and biotinylated complementary RNA (cRNA) from poly (A) RNA in a total RNA sample according to the user manual. IVT Incubation time was 16 hour. GeneAtlas® Hybridization, Wash and Stain Kit was used for hybridizing 3' IVT Array Strips according to the user manual (P/N 08-0306). Mouse genome array strips (MG-430) were hybridized for 16 hours in a 45oC incubator, washed and stained and finally imaging was done with the GeneAtlas Fluidics and Imaging Station.

基因表达谱分析(Gene expression profiling)揭示了木犀草素(Luteolin)在脂多糖(LPS)诱导抑郁模型中的潜在作用。本研究中,脂多糖诱导抑郁模型小鼠每日经口给予木犀草素(剂量为10 mg/kg体重),连续给药8天。对分离得到的小鼠神经干细胞(Neural Stem Cells, NSCs)开展基因芯片表达分析,每个实验使用2只小鼠的脑组织。本次基因芯片表达分析的样本包括:脂多糖诱导抑郁模型小鼠(LPS组)、经木犀草素处理的脂多糖诱导抑郁模型小鼠(LPS+L组)、正常小鼠(PBS组)以及经木犀草素处理的正常小鼠(PBS+L组)的神经干细胞与海马体组织。总RNA采用Isogen试剂(日本富山市日本基因有限公司)提取;采用NanoDrop 2000分光光度计(美国特拉华州威尔明顿赛默飞世尔科技公司)对RNA完整性进行定量检测。使用3'体外转录(IVT)Plus试剂盒(美国加利福尼亚州圣克拉拉市Affymetrix公司),将RNA样本制备为可用于GeneChip® 3'表达芯片的基因表达谱分析样本。按照试剂盒操作手册,每份样本取100 ng总RNA,从总RNA中的poly(A) RNA扩增并制备生物素标记的互补RNA(cRNA),体外转录孵育时长为16小时。按照操作手册(产品编号P/N 08-0306),使用GeneAtlas®杂交、洗涤与染色试剂盒完成3' IVT芯片条带的杂交实验。小鼠全基因组芯片条带(MG-430)在45℃孵箱中杂交16小时,随后完成洗涤与染色,最后通过GeneAtlas®流体力学与成像工作站完成成像扫描。
