This project proposes a combination of methods as an excellent solution to obtain low cost/large scale molecular data in biodiversity studies. We combined a especific platform and library to the genome skimming and obtained an innovative approach, opening a new range of possibilities of its use in obtaining molecular data from organisms with different levels of preservation. The principal advantages from our approach are: i. it requires low amount of input DNA (0.2 ng/ul), which allows the use of organisms with ancient DNA; ii. it does not depends on specific primers and is not affected by gene rearrangement; and iii. it is time-saving and cost-effective, leading to high-quality complete sequence assemblies.
本研究提出一种组合方法方案,可为生物多样性研究领域获取低成本、大规模分子数据提供优质解决方案。我们将特定平台与文库技术结合应用于基因组浅层测序(genome skimming),开发出一种创新性方法,为从不同保存状态的生物样本中获取分子数据拓展了全新应用可能。本方法的核心优势如下:其一,所需起始DNA量极低(0.2 ng/ul),可适用于带有古DNA的生物样本;其二,无需依赖特定引物,且不受基因重排影响;其三,省时高效且兼具成本效益,可获得高质量的完整序列组装结果。
