To decipher the T cell subset-specific consequences of IL-33 sensing, we profiled early changes in transcriptional activity of in vitro differentiated CTLs, Th1 cells and Th2 cells upon short-term stimulation with IL-33. Overall design: Comparative gene expression profiling analysis of the effects of IL-33 on CTLs, Th1 and Th2 cells using RNA-Seq and single-cell RNA-Seq of WT- and Il1rl1-ExAB-KO CTLs after an acute LCMV infection.
为解析白介素33(IL-33)感知所引发的T细胞亚群特异性生物学效应,我们对经IL-33短期刺激后的体外分化细胞毒性T淋巴细胞(Cytotoxic T Lymphocytes, CTLs)、Th1细胞及Th2细胞的转录活性早期变化开展了基因表达谱分析。实验整体设计:本研究采用RNA测序(RNA-Seq)与单细胞RNA测序(single-cell RNA-Seq)技术,对急性淋巴细胞性脉络丛脑膜炎病毒(Lymphocytic Choriomeningitis Virus, LCMV)感染后的野生型(Wild Type, WT)及Il1rl1-ExAB敲除(Il1rl1-ExAB-KO)细胞毒性T淋巴细胞进行测序,以此对比分析IL-33对CTLs、Th1细胞及Th2细胞的调控效应。
