Determine the direct effects of chromatin remodeler on transcription using inducible protein degradation combined with nascent transcriptomics. Overall design: Nascent RNA transcription measured by PRO-seq upon AID-mediated depletion of chromatin remodelers. All cell lines have OsTir1 expression from the Act5c locus, and AID tagged gene indicated in genotype. All experiments were performed in 2 biological replicates.
本研究采用诱导型蛋白质降解技术联合新生转录组学(nascent transcriptomics),旨在阐明染色质重塑因子(chromatin remodeler)对转录过程的直接调控效应。整体实验设计:通过PRO-seq检测染色质重塑因子经AID介导的降解后的新生RNA转录水平;所有细胞系均在Act5c基因座上表达OsTir1,且基因型中标注了带有AID标签的靶基因;所有实验均设置2次生物学重复。
