We compared transcriptomes of CD34(high)EPCR-positive with CD34(high)EPCR-negative cells after 10-days culture in PCL-PVAc-PEG based 3a medium. It was because our culture condition had not previously reported and so we demonstrated that phenotypic HSCs were concentrated in EPCR-positive fraction as conventional UM171-based culture system. As a result, CD34(high)EPCR-positive cells highly expressed HSC markers (HLF, AVP, PRDM16 and FGD5) compared with CD34(high)EPCR-negative cells. mRNA profile of CD34(high)EPCR-positive and CD34(high)EPCR-negative cells derived from 10-days cultured cord blood CD34+ cells.

本研究对比了在基于PCL-PVAc-PEG的3a培养基中培养10天后的CD34高表达且内皮细胞蛋白C受体(Endothelial Protein C Receptor, EPCR)阳性细胞与CD34高表达且EPCR阴性细胞的转录组。鉴于本研究所采用的培养条件此前未见报道,我们验证了与传统基于UM171的培养体系一致的结论:表型造血干细胞(Hematopoietic Stem Cell, HSC)富集于EPCR阳性细胞组分中。实验结果显示,相较于CD34高表达且EPCR阴性细胞,CD34高表达且EPCR阳性细胞高表达造血干细胞标志物(HLF、AVP、PRDM16及FGD5)。本数据集包含从10天培养的脐带血CD34阳性细胞中分离得到的CD34高表达且EPCR阳性与CD34高表达且EPCR阴性细胞的mRNA转录谱。
