To evaluate the robustness of CtG, we constructed Hi-C libraries of varying quantities of cell inputs utilizing a low-input Hi-C technique. Overall design: Both HEK293 and HEK293T cells were used. Either 1,000 cells or 10,000 cells were extracted to a 0.5 mL Eppendorf Lobind Microcentrifuge tube. Hi-C experiments were performed followed the method described in Rao et al., 2014 (PMID: 25497547) with some modifications.
为评估CtG的稳健性,我们采用低起始量高通量染色体构象捕获(Hi-C)技术,构建了不同起始细胞数量的Hi-C文库。整体实验设计:本研究选用HEK293与HEK293T两种细胞系,分别取1000个或10000个细胞转移至0.5 mL Eppendorf Lobind微量离心管中。随后参照Rao等人2014年发表的实验方案(PMID: 25497547)并进行部分优化,完成了全部Hi-C实验操作。
