We report the first discovery of naturally occurring ESR1Y537C and ESR1Y537S mutations in MCF7 and SUM44 ESR1-positive cell-lines after acquisition of resistance to long-term-estrogen-deprivation (LTED) and subsequent resistance to fulvestrant (ICIR). RNAseq examination of gene expression changes due to long term estrogen deprivation in two breast cancer cell lines with and without ESR1 mutations
本研究首次报道,在经长期雌激素剥夺(LTED)治疗后获得耐药性,并继而对氟维司群产生耐药(ICIR)的MCF7与SUM44雌激素受体1(ESR1)阳性细胞系中,自然发生了ESR1Y537C及ESR1Y537S突变。本研究同时针对两种分别携带与不携带ESR1突变的乳腺癌细胞系,开展了针对长期雌激素剥夺所致基因表达变化的RNA测序(RNAseq)分析。
