Purpose: The disco-interacting protein 2 homolog C (DIP2C) gene is an uncharacterized gene found mutated in breast and lung cancers. We want to understand the role of DIP2C in tumor development.Methods: We engineered human DIP2C knockout cell systems by genome editing, and then use next-generation sequencing to identify the genes affected by the loss of DIP2C.Results: Inactivation of DIP2C triggers substantial gene expression changes, cellular senescence and epithelial to mesenchymal transition in cancer cells.
研究目的:盘状结构域互作蛋白2同源物C(disco-interacting protein 2 homolog C,DIP2C)是一种尚未被功能表征的基因,其突变已在乳腺癌与肺癌样本中被检出。本研究旨在探究DIP2C在肿瘤发生发展过程中的作用。
研究方法:通过基因组编辑技术构建人源DIP2C敲除细胞系,随后采用下一代测序技术(next-generation sequencing)鉴定受DIP2C缺失影响的基因。
