We employed DNA microarray to figure out the possible mechanism of BBR on zebrafish NAFLD model Overall design: AB strain zebrafish (Danio. rerio) were kept under standard laboratory conditions with a 14-h light/10-h dark cycle at a temperature of 28.5±1°C. Embryos and larvae were maintained in embryo medium to 5 dpf without feeding. Zebrafish larvae were fed with control diet (AP100, 30mg/100 fish/day) or high cholesterol diet (HCD, 180mg AP100 plus 4% cholesterol/100fish/day) or HCD plus berberine (5μM) for 10 days.

本研究采用DNA微阵列(DNA microarray)技术,探究小檗碱(Berberine, BBR)对斑马鱼非酒精性脂肪性肝病(Non-Alcoholic Fatty Liver Disease, NAFLD)模型的潜在作用机制。实验设计:AB品系斑马鱼(Danio rerio)在标准实验室条件下饲养,光照周期为14小时光照/10小时黑暗,饲养温度维持在28.5±1℃。胚胎及幼鱼饲养于胚胎培养液中,至受精后5日龄(days post fertilization, dpf),期间不喂食。随后将斑马鱼幼鱼分为三组,分别投喂对照饲料(AP100,30mg/100尾鱼/天)、高胆固醇饲料(High Cholesterol Diet, HCD,180mg AP100添加4%胆固醇/100尾鱼/天),以及高胆固醇饲料联合5μM小檗碱,连续投喂10天。
