Transcriptomic analysis and identification of differentially expressed genes in wt vs KD Drosophila melanogaster ovaries. We compared gene expression profiles in Drosophila melanogaster ovaries in which the lid or the sin3A gene have been selectively knocked down by tissues specific shRNA expression. Overall design: Drosophila melanogaster ovaries mRNA profiles of 3-day old wild type (WT), germline specific lid knock-down (lidKD) germline specific sin3A knock-down female were generated by deep sequencing, in duplicate, using Illumina Hiseq 4000.

野生型(WT,wild type)与敲低型(KD,knockdown)黑腹果蝇(Drosophila melanogaster)卵巢的转录组分析及差异表达基因鉴定。本研究通过组织特异性短发夹RNA(shRNA,short hairpin RNA)的表达,分别对黑腹果蝇卵巢中的lid基因与sin3A基因进行选择性敲低,进而比较二者的基因表达谱。整体实验设计:采用Illumina Hiseq 4000平台,对3日龄野生型雌性果蝇、生殖系特异性lid敲低(lidKD)雌性果蝇以及生殖系特异性sin3A敲低雌性果蝇的卵巢mRNA转录组进行双重复深度测序。
