Ion torrent sequencing results of Atrogin-1 gene region among different chicken lines

To discover the SNPs information of Atrogin-1 gene region among chicken breeds, we targeted sequence 40 kb (chr 2: 143,690,694-143,730,638 bp) including upstream 10 kb and downstream 6 kb of this gene. RJF, Chahua, Tibetan, Silky, Fast Silky, mini, Cobb and AA were used (N = 50) (Chahua, Tibetan, Silky, Fast Silky and mini are Chinese native chicken breeds, while Cobb and AA are commercial broiler lines which derived from Cornish crosses). The PCR products were purified and subsequently sequenced by Ion torrent. Different chicken breeds were separated by barcode.

为解析不同鸡品种中Atrogin-1基因区域的单核苷酸多态性（Single Nucleotide Polymorphism，SNPs）信息，本研究针对该基因上下游共40 kb的序列区域（2号染色体：143,690,694-143,730,638 bp，其中上游延伸10 kb、下游延伸6 kb）开展靶向扩增。本研究共纳入8个鸡群，每组样本量为50（N=50），分别为原鸡（RJF）、茶花鸡、藏鸡、丝羽乌骨鸡、快大型丝羽乌骨鸡、矮小鸡、科宝（Cobb）肉鸡以及爱拔益加（AA）肉鸡；其中茶花鸡、藏鸡、丝羽乌骨鸡、快大型丝羽乌骨鸡及矮小鸡为中国地方鸡品种，科宝与爱拔益加肉鸡均为源自考尼斯鸡（Cornish）杂交培育的商业肉鸡品系。对聚合酶链式反应（Polymerase Chain Reaction，PCR）产物进行纯化后，采用Ion Torrent测序平台完成测序，通过Barcode（条码）区分不同鸡品种的测序序列。
