RNAseq was performed from Drosophila head tissue of two mitochondrial complex I deficiency models. The NDUFS1 homolog ND-75 was knocked down in Drosophila neurons using nSybGal4. Heads were collected from control of ND-75 knockdown flies and used for RNAseq analysis. Overall design: nSybGal4 flies were crossed to either w[1118] control flies or ND-75 RNAi flies (HMS00853). Heads were collected from 20 progeny for each genotype with 4 biological replicates.
本研究针对两种线粒体复合物I缺陷型果蝇模型的头部组织开展RNA测序(RNAseq)分析。采用神经元特异性Gal4驱动系nSybGal4,在果蝇神经元中敲低NDUFS1的同源基因ND-75。收集ND-75敲低组果蝇及其对照的头部组织,用于RNAseq分析。实验总体设计如下:将携带nSybGal4的果蝇分别与w[1118]对照果蝇及ND-75 RNA干扰(RNAi)果蝇(品系编号HMS00853)进行杂交。每个基因型的子代均收集20只果蝇的头部组织,设置4次生物学重复。
