TFAP2A (AP-2a) is a member of the transcription factor AP-2 family, functions as a tumor suppressor by regulating the expression of various cancer-related genes; its expression is positively correlated with chemosensitivity and survival of cancer patients. Overall design: SV-HUC-1 cells were infected with a lentivirus expressing scramble shRNA or AP-2a shRNA for 48 h and then the cells were selected with puromycin (1 µg/mL). Cells infected with scramble shRNA (control) and AP-2a shRNA were sequenced by Illumina HiSeq2000.
TFAP2A(AP-2α)属于转录因子AP-2家族成员,可作为抑癌因子,通过调控多种癌症相关基因的表达发挥功能;其表达水平与癌症患者的化疗敏感性及生存预后呈正相关。
整体实验设计:将SV-HUC-1细胞分别感染携带乱序短发夹RNA(short hairpin RNA,shRNA,阴性对照)或AP-2α shRNA的慢病毒,感染48小时后采用嘌呤霉素(1 μg/mL)进行筛选。随后分别对感染乱序shRNA的对照组细胞与感染AP-2α shRNA的实验组细胞进行Illumina HiSeq2000测序。
