The effects of aging on brain-associated T cells remain incompletely understood. We sorted CD4 and CD8 T cells from meninges and choroid plexus of young and aged mice, and compared their single-cell transcriptomes. The results provide insights into the changes of brain-associated T cells with aging, and identify unique aging-associated T cells. Meninges and choroid plexus samples were obtained from young and aged mice. CD4 and CD8 T cells were sorted by FACS. scRNA-seq were performed using the 10xgenomics 5' gene expression kit. Libraries were sequenced by a NextSeq.

衰老对脑相关T细胞的影响目前尚未完全阐明。本研究从年轻与衰老小鼠的脑膜(meninges)及脉络丛(choroid plexus)中分离分选CD4与CD8 T细胞,并对其单细胞转录组开展比较分析。研究结果为理解脑相关T细胞随衰老的动态变化提供了新的视角,并鉴定出一类独特的衰老相关T细胞。本研究的脑膜与脉络丛样本均取自年轻及衰老小鼠。CD4及CD8 T细胞通过荧光激活细胞分选(Fluorescence-Activated Cell Sorting, FACS)完成分选。采用10×基因组学(10x Genomics)5'端基因表达试剂盒完成单细胞RNA测序(single-cell RNA sequencing, scRNA-seq)实验。测序文库通过NextSeq测序平台完成测序。
