Transcriptome analysis of digital RNA-sequencing (dRNA-Seq) data reveals the differentially expressed genes between GFP-positive and GFP-negative cells. Overall design: GFP-positive cells and GFP-negative cells were sorted from the splenic human CD4-positive T cells of HIV1-GFP-infected humanized mice (n=6) at 2 weeks post-infection. The splenic human CD4-positive T cells were also sorted from mock-infected mice (n=7). By using these samples, dRNA-Seq was performed to performe transcriptome analysis (3 technical replicates for 3 infected mice and 3 mock-infected mice; 1 technical replicate for 3 infected mice and 4 mock-infected mice).
数字RNA测序(digital RNA-sequencing, dRNA-Seq)数据的转录组分析,揭示了绿色荧光蛋白(Green Fluorescent Protein, GFP)阳性与阴性细胞间的差异表达基因。
整体实验设计:于感染后2周,从HIV1-GFP感染的人源化小鼠(n=6)的脾脏人CD4阳性T细胞中分选得到绿色荧光蛋白阳性与阴性细胞;同时从模拟感染(mock-infected)小鼠(n=7)中分选得到脾脏人CD4阳性T细胞。利用上述样本开展dRNA-Seq测序以进行转录组分析:其中3只感染小鼠与3只模拟感染小鼠各设置3次技术重复;另外3只感染小鼠与4只模拟感染小鼠各设置1次技术重复。
