The goal of this project is to investigate the impact of circadian rhythm on resistance and tolerance. While work in some model insects (mainly D. melanogaster) has demonstrated a non-neutral effect of circadian rhythms on resistance to bacterial infection, the effect of circadian rhythms on disease resistance in agricultural pests, or tolerance in any organism, is unclear. Here we compare individuals that have been raised on a 12h:12h light:dark cycle to individuals that have been raised on the same cycle and transferred into dark conditions 24 hours prior to experimental start. Gene transcription associated with immunity, energy catabolism, and circadian rhythm will be compared.
本项目旨在探究昼夜节律(circadian rhythm)对宿主抗性与耐受性的影响。尽管已有针对部分模式昆虫(主要为黑腹果蝇(D. melanogaster))的研究证实,昼夜节律对细菌感染抗性存在非中性调控效应,但昼夜节律对农业害虫的病害抗性,或是对任意生物体的耐受性的影响仍不明晰。本研究将两组实验个体进行对比:一组在12小时光照:12小时黑暗的光暗周期环境中饲养,另一组在相同光暗周期饲养后,于实验开始前24小时转移至全黑暗环境中。后续将对与免疫、能量分解代谢及昼夜节律相关的基因转录水平开展比较分析。
