Hormone drugs can relieve pain in endometriosis patients but cannot cure the disease, so curative therapy for endometriosis is an urgent unmet medical need. We found that the progression of endometriosis is associated with inflammation and fibrosis. In addition, IL-8 expression was highly upregulated in endometriotic tissues and closely correlated with disease progression. To develop a disease-modifying drug for endometriosis, we created a long-acting recycling antibody against IL-8 and evaluated its clinical potency in monkeys with spontaneously developed or surgically induced endometriosis. As part of the evaluation, we performed the fast gene set enrichiment analysis and upstream regulator analysis(Ingenuity pathway analysis) with RNA sequencing data from monkey with spontaneouly developled endometriosis before and after anti-IL8 antibody treatment.
激素药物可缓解子宫内膜异位症(endometriosis)患者的疼痛,但无法根治该疾病,因此子宫内膜异位症的根治性治疗仍是亟待满足的医疗需求。本研究发现,子宫内膜异位症的进展与炎症及纤维化密切相关。此外,白细胞介素8(IL-8)在异位子宫内膜组织中呈高表达状态,且与疾病进展紧密相关。为开发针对子宫内膜异位症的疾病修饰药物,本研究制备了一款靶向IL-8的长效循环抗体,并在自发形成或手术诱导子宫内膜异位症的猕猴中评估其临床效力。作为评估的一部分,本研究利用自发形成子宫内膜异位症猕猴在接受抗IL-8抗体治疗前后的RNA测序数据,开展了快速基因集富集分析及上游调控因子分析(Ingenuity通路分析,Ingenuity pathway analysis)。
