本研究检测经或未经I-PpoI酶切、转染阴性对照（NC）或靶向METTL3的小干扰RNA（siMETTL3）的HEK293T细胞中，I-PpoI酶切位点（28S核糖体DNA（28S rDNA）与DAB1基因）及其他未酶切基因组区域的R环（R-loop）形成情况。总体实验设计：采用S9.6抗体从上述处理的细胞中分离R环，随后在HiseqXten测序平台进行双端测序。
