To investigate the potential pathogenic mechanism of glioma-related epilepsy (GRE), we have employed analyzing of the dynamic expression profiles of microRNA/ mRNA/ lncRNA in brain tissues of glioma patients. Brain tissues of 16 patients with GRE and nine patients with glioma without epilepsy (GNE) were collected. The total RNA was dephosphorylated, labeled, and hybridized to the Agilent Human miRNA Microarray, Release 19.0, 8x60K. The cDNA was labeled and hybridized to the Agilent LncRNA+mRNA Human Gene Expression Microarray V3.0, 4x180K. The raw data was extracted from hybridized images using Agilent Feature Extraction, and quantile normalization was performed using the Agilent GeneSpring. We found that three differentially expressed miRNAs (miR-10a-5p, miR-10b-5p, miR-629-3p), six differentially expressed lncRNAs (TTN-AS1, LINC00641, SNHG14, LINC00894, SNHG1, OIP5-AS1), and 49 differentially expressed mRNAs may play a vitally critical role in developing GRE. Overall design: Brain tissues of 25 glioma patients with or without epilepsy were retrospectively obtained from the Affiliated Cancer Hospital of Xiangya School of Medicine (Changsha, Hunan, China) with informed consent.

为探究胶质瘤相关性癫痫(glioma-related epilepsy, GRE)的潜在致病机制,本研究通过分析胶质瘤患者脑组织中微小RNA(microRNA, miRNA)、信使RNA(messenger RNA, mRNA)与长链非编码RNA(long non-coding RNA, lncRNA)的动态表达谱开展相关研究。本研究共收集16例GRE患者与9例无癫痫胶质瘤(glioma without epilepsy, GNE)患者的脑组织标本。对总RNA进行去磷酸化、标记处理后,将其与安捷伦人类miRNA微阵列(Agilent Human miRNA Microarray)Release 19.0、8x60K进行杂交;对cDNA进行标记后,与安捷伦LncRNA+mRNA人类基因表达微阵列V3.0、4x180K进行杂交。原始数据通过安捷伦特征提取软件(Agilent Feature Extraction)从杂交图像中提取,并采用安捷伦GeneSpring软件进行分位数归一化处理。本研究发现,3种差异表达miRNA(miR-10a-5p、miR-10b-5p、miR-629-3p)、6种差异表达lncRNA(TTN-AS1、LINC00641、SNHG14、LINC00894、SNHG1、OIP5-AS1)以及49种差异表达mRNA可能在GRE的发生发展中发挥至关重要的作用。总体实验设计:本研究回顾性获取了25例伴或不伴癫痫的胶质瘤患者的脑组织标本,所有标本均来自中国湖南省长沙市湘雅医学院附属肿瘤医院,且已获得患者知情同意。
