Plasmodium falciparum transgenic parasite lines and ChIP-seq transcription factor AP2-EXP

In this study we have generated transgenic parasite lines to endogenously tag the ApiAP2 protein AP2-EXP. We performed ChIP-seq to measure AP2-EXP genomic occupancy in the parasite lines AP2-EXP::GFP and AP2-EXP::HA at 30 hours post invasion. A negative control ChIP-seq experiment was done on wildtype 3D7 P. falciparum.

本研究构建了对内源ApiAP2蛋白AP2-EXP进行标记的转基因寄生虫品系。我们采用染色质免疫共沉淀测序（ChIP-seq）技术，在入侵后30小时的AP2-EXP::GFP与AP2-EXP::HA寄生虫品系中检测AP2-EXP的全基因组结合分布。同时以野生型3D7恶性疟原虫（P. falciparum）为材料开展了ChIP-seq阴性对照实验。
