To demonstrate the role of miRNAs in desmoplastic stroma of pancreatic cancer patients sample, we compared the miRNA expression patterns in tumor, stroma and normal tissue. Total RNA was extracted from formalin fixed paraffin embedded samples from the pancreatic cancer patients samples. The miRNA expression profiling is done on tumor, stroma and normal tissues seperately using miRCURY LNATM microRNA Array 7'th arrays (Exiqon, Denmark). The expression profiles were background corrected and quantile normalized. Filtering of controls and low expressed probes was done. 755 miRNAs were used to demonstarte the role of miRNAs in tumor and stroma interaction and in carcinogenesis.

为阐明微小RNA(microRNA,miRNA)在胰腺癌患者样本促结缔组织增生性间质中的作用,本研究对比了肿瘤组织、间质组织与正常组织的miRNA表达谱。总RNA提取自胰腺癌患者的福尔马林固定石蜡包埋样本;本研究采用丹麦Exiqon公司的miRCURY LNATM 第7代微小RNA芯片,分别对肿瘤组织、间质组织及正常组织进行miRNA表达谱分析。对所得表达谱进行背景校正与分位数归一化处理,并完成对照探针与低表达探针的筛选,最终选取755个miRNA,用于阐明miRNA在肿瘤与间质相互作用及癌变进程中的作用。
