We first found that Cyclin-Dependent Kinase Like 3 (CDKL3) plays an important role in Non-Alcoholic Fatty Liver Disease (NAFLD). Using the Cre-Loxp system, we constructed the murine model with a liver-specific Cdkl3 deletion and then comprehensively evaluated the effects of liver Cdkl3 deficiency through high-throughput sequencing. After knocking out Cdkl3 in the liver (C57BL/6), male mice (control and knockout group) were selected and fed with a chow diet for 10 months, followed by sequencing transcriptome profiling of whole livers.

本研究首先发现周期蛋白依赖性激酶样3(Cyclin-Dependent Kinase Like 3,CDKL3)在非酒精性脂肪性肝病(Non-Alcoholic Fatty Liver Disease,NAFLD)中发挥关键作用。我们借助Cre-Loxp重组酶系统(Cre-Loxp system)构建了肝脏特异性敲除Cdkl3的小鼠模型,并通过高通量测序技术全面评估肝脏Cdkl3缺失所产生的生物学效应。在C57BL/6小鼠的肝脏中敲除Cdkl3后,选取雄性小鼠分为对照组与敲除组,以普通饲料饲喂10个月,随后对其全肝组织开展转录组测序分析。
