We present a chemical method (m6A-ORL-Seq) for transcriptome-wide m6A profiling , and compared its results with MeRIP-seq. Overall design: Transcriptome-wide Mapping of N6-Methyladenosine on HEK-293T by a chemical method (m6A-ORL) and an antibody-based method (MeRIP-seq), then analyzed and compared the performance of those two methods. M6A-ORL aslo provides four thousands of high-confidence m6A sites based on chemical modified m6A induced RT truncation.
本研究报道了一种可实现转录组范围N6-甲基腺嘌呤(m6A)图谱分析的化学方法(m6A-ORL-Seq),并将其检测结果与MeRIP-seq进行了对比。
实验设计概况:在HEK-293T细胞中开展转录组范围N6-甲基腺嘌呤定位分析,分别采用化学方法(m6A-ORL)与基于抗体的方法(MeRIP-seq),随后对两种方法的检测性能进行分析与对比。
此外,基于化学修饰的m6A诱导逆转录(Reverse Transcription, RT)截短信号,m6A-ORL方法还可得到四千余个高置信度的m6A位点。
