In this study we sequenced RNA of granulosa collected from Holstein cows at oocyte aspiration prior to IVP, to identify candidate genes and biological mechanisms for favourable IVP-related traits in donor cows. Overall design: 24 RNA-Seq profiles of granulosa cells collected from antral follicles of 24 cows. Each animal was scored for In Vitro Production performances (blastocysts rate, morphology and kinetic scores). Half of the oocytes were inseminated with semen from high genetic merit bulls and half with semen from low genetic merit bulls.
本研究对荷斯坦奶牛在体外生产(In Vitro Production, IVP)前进行卵母细胞抽吸时采集的颗粒细胞(granulosa cells)的RNA进行测序,旨在筛选供体奶牛中与优良IVP相关性状相关的候选基因及生物学机制。
总体设计:从24头奶牛的窦状卵泡(antral follicles)中采集颗粒细胞,共计获得24份RNA测序(RNA-Seq)转录组谱。对每头奶牛的体外生产性能进行评分,评分指标涵盖囊胚率、胚胎形态及发育动力学评分。实验中,一半卵母细胞采用高遗传品质公牛的精液进行授精,另一半采用低遗传品质公牛的精液授精。
